Eines schönen Tages hört der neugierige Elefant Horton im Dschungel einen Hilfeschrei von einem Staubkorn. Auf ihm wohnen die Hus, deren winzige Welt in Gefahr ist. Horton verspricht den Winzlingen, sie wieder an einen sicheren Platz im Dschungel zu bringen. Dieses Versprechen katapultiert ihn mitten in ein rasantes Dschungelabenteuer. Die anderen Urwaldbewohner, allen voran die garstige Känguru-Mama, halten ihn nicht nur für vollkommen verrückt, sondern versuchen sogar, seine Mission zu verhindern.
